10-F05 is a covalent small molecule inhibitor of bacterial β-ketoacyl-acyl carrier protein synthase III (FabH) and MiaA tRNA prenyltransferase, demonstrates broad-spectrum antibacterial effectiveness (MIC=2.5 uM, S. flexneri).
10-F05 is effective against multiple ESKAPE pathogens and demonstrates a slow resistance development.
10-F05 exhibits low cytotoxicity in HEK293T cells.
10-F05 specifically reacts with MiaA Cys273, disrupts MiaA and tRNA substrate binding.
10-F05 reduces translation fidelity and stress resistance through MiaA inhibition.
