(S)-STO021 is a potent, selective agonist of estrogen receptor β (ERβ) with EC50 of 1.4 uM, >70-fold selective over ERα.
(S)-STO021 (1 μM) promoted osteogenic differentiation through the ERβ/BMP4/SMAD signaling pathway.
(S)-STO021 inhibited osteoclast differentiation through the ERβ/TAK1/MKK6/p38 signaling pathway.
STO021 could not only effectively improve the zebrafish osteoporosis model induced by glucocorticoids but also greatly restore the bone microstructure in ovariectomized rat models.
